An analysis of phagocyte subpopulations will be performed using cryopreserved lung cells, peripheral blood, and biopsies from all available study samples. In addition, functional immunological tests, phagosome maturation studies, and cytokine production studies will be performed after infection with pathogens (as part of work package 6). In detail: 1. Lung cells will be infected with fungus and then stained with special fluorescent dyes and analyzed using confocal microscopy. 2. Stimulation with macrophage fungal spores will be performed, followed by analysis of phagosome maturation after staining with specific antibodies for marker protein labeling. 3. The ability of macrophages to inhibit the growth of pathogenic fungi will be evaluated. 4. Differentiation and stimulation of macrophages from peripheral blood monocytes will be performed. 5. Dynamic phagosome maturation tests in live cells and after stimulation and fixation will be thoroughly analyzed using microscopy techniques (as part of work package 6 of the project).
