Project: “Personalized antibody-drug conjugates activated by matrix metalloproteinases for solid tumors”, acronym: PACMAN, Virtual Research Institute (WIB); managing entity: Łukasiewicz Research Network – PORT. The PACMAN project aims to develop a new class of antibody-drug conjugates (ADCs) that do not require internalization and are selectively activated extracellularly by matrix metalloproteinases (MMP-2, MMP-9, MMP-14) in the solid tumor microenvironment.

Employment period and remuneration: 12 months (with the possibility of extending the employment for an additional 36 months following a positive performance evaluation, for a total employment period of 48 months); monthly salary 12 000 gross/gross.

Job description: Research Specialist in the Research Group led by Prof. Agnieszka Dobrzyń.

Tasks: Work on the implementation of the project in the area of in vitro and in vivo testing of newly developed antibody–drug conjugates (ADCs) for pancreatic cancer.

 

Requirements:

- Applicant must hold a PhD degree in Biological Sciences or a closely related discipline and demonstrate documented hands-on experience with the independent application of laboratory techniques in molecular biology, cell biology, biochemistry, and histology. Required qualifications include experience in DNA and RNA extraction and purification, plasmid isolation, PCR and RT-qPCR, as well as agarose gel electrophoresis. The candidate should also demonstrate proficiency in protein extraction and quantification from tissue and cell samples, SDS-PAGE electrophoresis, Western blotting, and enzyme activity assays. Practical experience with light, fluorescence, and confocal microscopy is required.

- Applicant should have experience in mammalian cell culture, including the maintenance of primary cell cultures. Familiarity with cell viability and proliferation assays (Neutral Red, MTT, and MTS), assessment of cellular redox status using Griess, DPPH, and ferric reducing assays, as well as immunocytochemistry and cytochemical staining techniques is required.

- The candidate should have experience in tissue collection, tissue freezing and cryosectioning, histological staining techniques including Hematoxylin and Eosin (H&E), Oil Red O, and Picro-Sirius Red staining, as well as immunohistochemistry. Experience in handling and monitoring laboratory mice, performing euthanasia procedures in accordance with ethical guidelines, and conducting intramuscular cardiotoxin injections under anesthesia is also required.

- Experience with advanced research methodologies, including Chromatin Immunoprecipitation Sequencing (ChIP-Seq), CRISPR/Cas9 genome editing, Seahorse metabolic analysis, and zymography, will be considered an advantage.

- Complying with the non-competition policy following the WIB Programme guidelines is required.

1. The candidate’s scientific and professional experience, as well as their competencies relevant to the implementation of tasks within the research project.

2. Scope of previous scientific activity.

3. Quality and scope of the candidate’s scientific achievements, in particular scientific publications, participation in research projects, awards and distinctions received, and references provided by independent researchers.

4. The candidate’s soft skills, including the ability to work effectively in a team, communication skills, proficiency in English, and the alignment of the candidate’s scientific development plans with the objectives of the research group.
