Name of research project: CHIMERNA - Cancer therapy Hub based on smart nanoparticles for in situ IMmune cell Engineering with mRNA 

 The aim of the project will be to develop a hybrid, hydrogel-based biomaterial that will promote the formation of a lymph node-like structure. This approach will aim to recruit antigen-presenting cells and program them, by transfection with mRNA, to present tumor antigens to CD4 and CD8 T cells, thereby enhancing the efficacy of the immune response against cancer.

Activity 1: Optimize lipid nanoparticle-based hydrogels for sustained chemokine and mRNA release.

- Task 1.1: formulate various lipid NPs, characterize them, verify interactions with chemokine XCL1 and a GFP mRNA.

- Task 1.2: formulate optimized lipid NP hydrogels, characterize them, and verify that interactions with chemokine XCL1 and a GFP mRNA are still intact.

- Task 1.3: study the release profiles of biological molecules and the possibility of syringing hydrogels 

Activity 2: Chemotaxis and transfection of cDC1 dendritic cells using hydrogels

- Task 2.1: Verify the chemotaxis of mouse cDC1 in vitro and in vivo.

- Task 2.2: verify transfection of migrating cDC1s with released GFP mRNA, and its impact on cDC1 phenotype.

Activity 3: Follow-up of the CHIMERNA project.

- Task 3.1: presentation of results at meetings with the consortium and drafting of deliverables.

- Task 3.2: management of consumables orders and co-direction of students involved in the classroom project
