- work environment: Of the ~40 known modifications in mammalian tRNAs, queuosine (Q) is unique in its dependence on an external metabolite from the gut microbiome or diet. The molecular pathway involves the incorporation of queuosine (q), a microbial micronutrient, into the position of the wobble anticodon (G34) of tRNATyr, tRNAHis, tRNAAsn and tRNAAsp (collectively referred to as tRNAs-Q), thereby regulating the efficiency and fidelity of mRNA translation. As such, tRNAs-Q modification offers a direct means for the microbiome to influence host gene expression and cellular physiology. This multidisciplinary project aims to take advantage of the knowledge gained from studying host-microbiome interactions and RNA chemistry to design unique synthetic molecules capable of blocking protein synthesis and reducing the growth of cancer cells.

- main mission: Test the impact of bacterial metabolites and a synthetic derivative on tRNA metabolism, gene expression and tumour growth, in vitro and in vivo.

- Extract and purify tRNAs

- Detect and quantify the incorporation of metabolites into RNA by LC-MS/MS

- Study the stability of targeted tRNAs by Northern Blot

- Study the impact on gene expression (transcriptome, translatome)

- Study the phenotypic consequences on healthy and cancerous cells, in vitro and in vivo

- Identify the molecular pathways involved by gene inactivation
