Project Description

This position is funded under project CPP2024‑011546 (ARA) and is hosted at the University of Barcelona, within the research line Molecular Oncology and Embryology. The project aims to develop and validate a novel non‑invasive preimplantation genomic test based on the analysis of genomic DNA enclosed in extracellular vesicles (EVs) present in human embryo culture medium.

By isolating and characterizing embryo‑derived EVs and analysing their genomic cargo, the project seeks to overcome current limitations of non‑invasive PGT‑A and to provide a more accurate, reliable and embryo‑safe genomic screening strategy that can be applied in assisted reproduction clinics. The project integrates expertise from public and private partners and contributes to advancing reproductive genetics and embryology.


Responsibilities and Functions of the Position

The postdoctoral researcher will carry out activities directly linked to the development, optimisation and validation of the new EV‑based non‑invasive genomic test, as well as complementary tasks within the group’s research line. The main responsibilities include:


1. Optimisation and validation of extracellular vesicle (EV) isolation protocols

  • Implement and compare isolation strategies such as ultracentrifugation, size‑exclusion chromatography (SEC), polymer‑based precipitation, immunocapture and density gradients.
  • Establish a robust, scalable and clinically transferable EV‑isolation workflow.

2. Analysis of genomic DNA (gDNA) contained in EVs

  • Purify ultra‑low‑input gDNA from isolated EVs.
  • Evaluate DNA concentration, integrity and chromosomal representativeness.
  • • Explore complementary molecular markers, including global DNA methylation.


    3. Adaptation of niPGT‑A protocols to the EV‑based approach

  • Integrate exosomal gDNA into MALBAC®‑based whole‑genome amplification workflows.
  • • Support the optimisation of amplification and indexing steps feeding into NGS library preparation.


    4. Technical coordination with consortium partners

  • Coordinate sample transfer, controls and data exchange for sequencing.
  • Contribute to the evaluation of data quality generated by partners.
  • Support comparative validation and propose improvements in isolation and amplification procedures.


  • 5. Development of complementary methodological approaches
  • Evaluate methylation‑based strategies to confirm the embryonic origin of DNA.
  • Explore small and long RNAs in EVs as potential biomarkers of embryo competence.
  • Propose methodological enhancements based on preliminary results.


  • 6. Scientific coordination and consortium engagement
  • Participate in periodic meetings and preparation of progress reports.
  • Draft protocols, technical documents, project reports and patent‑related material.
  • • Contribute to manuscripts and international conference communications.


    7. Additional assigned tasks: development and management of 3D models (organoids)

  • Support the development, refinement and maintenance of embryonic and tumour‑derived organoid models.
  • Perform morphological and molecular characterisation (microscopy, PCR, differentiation markers).
  • Contribute to studies on developmental mechanisms and EV–cell interactions.
  • • Integrate organoid systems as potential complementary validation platforms.

    Gross salary per year: 24.443,88 €.

  • The application period is from March 16 to March 27, 2026, both inclusive.
