The project will be conducted within the “Translation Mechanisms” team led by Emmanuelle Schmitt and Yves Mechulam at the “Structural Cell Biology” (BIOC) laboratory at École Polytechnique, https://portail.polytechnique.edu/bioc/en/recherche/translation-mechani…. The BIOC laboratory is a joint research unit of the CNRS and École Polytechnique, located on the Palaiseau campus and fully equipped to carry out the project. In addition, the laboratory has privileged access to CIMEX (Interdisciplinary Center for Electron Microscopy at the École Polytechnique), which is equipped with a 300 kV Titan Themis electron microscope featuring a Falcon 3 direct electron detector.

Our results establish that HibA is a key mediator of ribosomal hibernation in archaea. They provide a mechanistic and functional framework for understanding how archaeal cells preserve their translation apparatus under stress, and open up new perspectives on the evolution and regulation of ribosomal dormancy across the three domains of life. This thesis project builds on our previous work (PMID: 42045235) and aims to further investigate the mechanisms of hibernation in archaea.
(i) To track the assembly of ribosomal subunits in the presence of HibA, we propose to conduct pre-steady-state kinetic experiments using a stopped-flow apparatus. Using this method, we will be able to determine the association rate constant (kon) of the 30S and 50S subunits and measure the influence of HibA on this association kinetics. The influence of ATP or AMP on this association rate will also be measured, as well as that of competitors (mRNA and/or transfer RNA, or the ABCE1 recycling factor).
(2) SBDS has been identified as a potential partner of HibA. Thus, another objective will be to clarify the function of the SBDS protein in archaea, beginning with determining whether aSBDS is involved in the final stages of biogenesis.
(3) Other partners of HibA will also be investigated.
