Androgen and Glucocorticoid Receptors Cross-activities and Oligomerization – ANAGRAM.

Research Background. Our preliminary resuts shows the formation of an androgen-glucocorticoid receptors heterodimer, AR·GR, which is the cornerstone of this project.

Overall functions of the candidate include solving the molecular determinants of an AR·GR complex Orthogonal biophysical and biochemical complementary techniques will be used in parallel.

Concrete tasks and responsabilities:

To determine binding and kinetic constants of peptides from AR and GR and protein domains in solution by T200 Surface plasmon resonance.
How do these constants compare with those of the respective homodimers and atypical nuclear receptor heterodimers.
To determine the binding and kinetic constants of the GR- LBD·AR-LBD heterodimer as well as of the AR·GR cores in the presence of small- molecule drugs usend in the clinic in AR and GR-related pathologies.
To characterize complex formation in living cells by Number and Brightness and Single Molecule Tracking.
To anallize complex formation by protein-protein docking and cutting edge AI methods in silico.

Skills/Qualifications

Physics X-ray diffraction mathematics, X-ray crystallography, diffraction data indexing, scaling, refinement, CCP4, COOT, Pymol, molecular replacement, surface plasmon resonance, protein chemistry, crosslinking, MS-spectrometry, SDS-PAGE, AI in protein structure.

Specific Requirements

To have 20 or more years experience in X-ray crystallography and Physics background and be expert in X-ray crystallography, diffraction data indexing, scaling, refinement. To have been in different highly competitive international institutes around the world not only Europe. To have prior experience leading students and having published impactful papers in the structural biology field. Required to have a high command English and German.
