The recruited person will carry out the research related to the ANR HoloFungi project that aim at studying the chemical interaction that occur within a toxic dinoflagellate holobiont with a specific focus on fungi. This is part of an ongoing project that already provides preliminary results published in Environ. Microbiol. 2023, 25, 250-267, (doi: 10.1111/1462-2920.16271).

The recruited person will be involved in different part of the project that will be carried out in parrel according to partners achievements and availability.

The first part of the project, in close collaboration with the Toulouse Biotechnology Institute, will explore the nutrient exchange between the dinoflagellate and fungi through a fluxomic approach using stable isotopic labelling. Kinetic measurements of metabolites in the extracellular medium will be achieved using ¹H-NMR (proton nuclear magnetic resonance) methodology. The recruited person will be in charge of micro-organisms culture with isotope labelling, extraction, profiling and data analysis.

The second part of the project, in close collaboration with Ifremer Nantes, will explore the chemical mediation between the dinoflagellate and fungi through a metabolomic approach (using liquid chromatography coupled to high resolution mass spectrometry – LC-HRMS). This part of the project will focus on induced chemical novelty along with the impact of microbial co-culture on toxin production. The recruited person will be in charge of micro-organisms culture, extraction, profiling and data analysis.

The last part of the project will be the isolation of compounds produced by fungal strains isolated from the toxic dinoflagellate through traditional natural products chemistry strategy. After large scale extract production, compounds will be isolated by flash and high-performance liquid chromatography. Pure compounds will be characterised (1D and 2D NMR and high-resolution MS) prior to their biological evaluation on the dinoflagellate and/or as potential anti-microbials.

In addition to the previously described activities, the recruited person will be in charge of (1) sample and data management between partners, (2) result valorisation through scientific publication, presentation in scientific conferences and also to non-scientific audiences.

Salary : € 2.800 gross per month
