The project aims to develop a system to produce posttranslational modified proteins and use them for functional and biophysical studies. Specifically, we want to understand how modifications of proteins change their propensity for misfolding and amyloid formation. 

The lab of Kristine Steen Jensen is looking for two highly motivated and self-driven postdoctoral research-candidates; one developing a system for producing proteins with posttranslational modifications (PTMs) through expansion of the genetic code (project I) and one focusing on functional and biophysical characterization of modified proteins (project II).


Job description:

Project I has a biochemical focus and the candidate is expected to develop a high throughput assay to study the coupling between local codon usage and expression yields of proteins with noncanonical amino acids. The work will include generation of DNA libraries, development of cell-free expression systems for screening of incorporation of noncanonical amino acids, fluorescent based quantification of expression levels and development of data analysis strategies for the high throughput data generated in the project. 

Project II has a biophysical focus and the candidate is expected to conduct biophysical characterization of amyloid forming reactions to outline effects of posttranslational modifications protein structure, function and dynamics. A combination of techniques will be applied including NMR spectroscopy, optical spectroscopy, electron microscopy and scattering methods. The candidate is expected to express and purify protein in collaboration with the postdoc on project I.


Where?

The researchers will join KS Jensen research group that studies the biophysical characterizations of the impact of posttranslational modifications on protein structure, function and dynamics. We are part of Division for Biophysical Chemistry, Center for Molecular Protein Science (https://www.cmps.lu.se), Department of Chemistry, Lund University, Sweden. Ongoing research at the Division of Biophysical Chemistry includes studies of biomolecular interactions, structure, and dynamics, aiming to understand biological systems and processes at the molecular level. The focus is on experimental and theoretical studies of the physical properties of proteins in solution. We have state of the art facilities for protein chemistry and protein NMR spectroscopy. 


Qualifications:


Project I: The candidate should hold a PhD or equivalent degree in a relevant field such as biochemistry or molecular biology. The candidate should be self-motivated and independent, be collaborative, and be interested in contributing to the common good of the lab. Good skills in oral and written English are required.

The candidate must have experience in molecular studies applied to biological macromolecules. Previous experience with flow cytometry or FACS is beneficial. An interest in automation of experimental work using robotic setups is also beneficial.


Project II: The candidate should hold a PhD or equivalent degree in a relevant field such as biochemistry, Biophysical chemistry or molecular biology. The candidate should be self-motivated and independent, be collaborative, and be interested in contributing to the common good of the lab. Good skills in oral and written English are required.

The candidate must have experience in molecular studies of proteins. The candidate should have experience and skills in biophysical characterization of protein structure and dynamics. Specifically, experience with biomolecular NMR is considered an advantage.
