- work environment: 

The Centre de Recherche en Biologie Cellulaire de Montpellier (CRBM) is one of the 3 research units attached to the Biology division on the CNRS route de Mende site in Montpellier. It is home to 160 people, 19 research teams, an R&D team, a start-up and the CRBM-MRI platform of the UAR BioCampus. The CRBM has made a name for itself in particular through its “cell cycle” theme, which has earned the laboratory international renown thanks to the discovery of the molecular mechanisms that control cell division. Today, the CRBM develops fundamental research specialized in cell biology. This recruitment aims to complement the experimental approach of the “Cytoplasmic Control of Genome Stability” research team, where the Saccharomyces cerevisiae and human cell study models are already well established, by now consolidating a new study model, namely the tardigrade Hypsibius exemplaris.

- main mission: 

The question to be answered is what is the response of lipid droplets (content, size, proteome) in response to DNA damage, and what is the fate of triacylglycerols previously stored in such lipid droplets: are they excreted? by what route?

The chosen researcher must have expertise in the culture, manipulation and analysis of tardigrades. The person will be expected to develop molecular biology, cell biology and microscopy techniques in this animal, such as immunofluorescence, genomic and protein DNA extraction, FISH, metabolic labeling with EdU and EU, RNA extraction for sequencing, lipidomic and proteomic analyses, in relationship with the above mentioned biological questions, ensuring their transmission and correct execution/application within the team.

- activities: 

- Culture, manipulation and data analysis of the tardigrade model .

- Implement genetic modification strategies in the model (shRNA, CRISPR-Cas9).

- Develop protocols for microscopic monitoring (in line with the options available on the on-site MRI platform) of the cellular processes studied by the team. In situ hybridization, immunofluorescence, cloning and protein labeling for microscopy of live samples will be practiced on a recurrent basis.

- Coordinate with the electron microscopy platform to optimize sample preparation.

- Preparation of high-quality RNA/DNA samples for sequencing and subsequent analysis.

- Philogenetic analysis of genes, proteins and domains (especially those involved in lipid binding).

- Preparation of high-quality lipid droplet samples for proteomics and lipidomics and subsequent analysis.

- Image processing and analysis

- Statistical analysis of data

- Processing and interpreting results

- Formatting results and interpreting them (for oral presentations, public dissemination, congresses, publication (writing of scientific articles), as well as, eventually, patents.

- Present results at seminars and conferences

- Draft funding applications
